Background

Our specialist in aseptic handling was tasked with leading a critical fast-track project aimed at scaling up seeding and media preparation within a pharmaceutical production facility. This initiative was essential to address the increasing demand for aseptic operations, requiring rigorous planning and execution to ensure compliance with industry standards and regulatory requirements.

Challenges

The primary challenge revolved around the project’s tight deadline, necessitating the fulfillment of all Good Manufacturing Practice (GMP) requirements and compliance with ANNEX 1. Simultaneously, the seamless transition of seeding and media preparation processes to an aseptic isolator environment had to be achieved without compromising the integrity of the products.

Approach

Kasper initiated the project by conducting a comprehensive durability study using mock-ups to assess the feasibility of performing all working procedures within the aseptic isolator. This step was crucial in evaluating the isolator’s design and functionality.

Upon confirming that the procedures could be effectively handled within the aseptic isolator, Kasper proceeded to prepare the User Requirement Specification (URS), Engineering Specification, and tender materials. These documents formed the foundation for outlining specific requirements, technical specifications, and procurement needs essential for the project’s successful implementation.

 
Subsequently, a comprehensive Design Qualification (DQ) was performed to ensure that all requirements outlined in the URS and Engineering Specification were met in the design of the aseptic isolator. Additionally, measures were taken to ensure traceability of all requirements to the Factory Acceptance Test (FAT) and Site Acceptance Test (SAT).

Outcome

Kasper’s diligent efforts resulted in the successful execution of the aseptic scale-up project for seeding and media preparation. The project not only adhered to predefined timelines and budget constraints but also ensured unwavering compliance with all regulatory and quality standards throughout the process.
